What is a TaqMan assay?
TaqMan genotyping assays are used to amplify and detect specific alleles in genomic DNA (gDNA). The exonuclease activity of AmpliTaq Gold® DNA Polymerase cleaves only probes hybridized to the target. Cleavage separates the reporter dye from the quencher dye, increasing fluorescence by the reporter.

Applied Biosystems TaqMan Gene Expression assays are used for quantitative real-time PCR analysis of gene expression and consist of a pair of unlabeled PCR primers and a TaqMan probe with a dye label (FAM) on the 5′ end and a minor groove binder (MGB) and non-fluorescent quencher (NFQ) on the 3′ end.
What does a quencher do?
In materials science, quenching is the rapid cooling of a workpiece in water, oil or air to obtain certain material properties. A type of heat treating, quenching prevents undesired low-temperature processes, such as phase transformations, from occurring.
What is TaqMan master mix?
TaqMan™ Master Mixes contain buffer, dNTPs, passive reference dye, thermostable hot-start DNA polymerase, and other components formulated for reliable TaqMan™ Assay-based real-time PCR. Just add your sample and TaqMan Assay, and start your reactions.
Is Tamra a quencher?
TAMRA dye is an effective quencher for fluorophores with emission maxima less than 560 nm. Dyes with longer wavelength emissions will not be effectively quenched by TAMRA. In addition, TAMRA has its own fluorescence which complicates data analysis due to crosstalk between the channels.
Why is fluorescent dye used in qPCR?
Dye-based quantitative PCR (qPCR) uses real-time fluorescence of a double-stranded DNA (dsDNA) binding dye (e.g., SYBR® Green) to measure DNA amplification as it occurs during PCR. The dye displays weak background fluorescence that increases dramatically upon binding to dsDNA.
